Nucleoside transporter-guided cytarabine-conjugated liposomes for intracellular methotrexate delivery and cooperative choriocarcinoma therapy

Abstract Gestational trophoblastic tumors seriously endanger child productive needs and the health of women in childbearing age. Nanodrug-based therapy mediated by transporters provides a novel strategy for treatment tumors. Focusing on overexpression human equilibrative nucleoside transporter 1 (ENT1) membrane choriocarcinoma cells (JEG-3), cytarabine (Cy, substrate ENT1)-grafted liposomes (Cy-Lipo) were introduced targeted delivery methotrexate (Cy-Lipo@MTX) this study. ENT1 has high affinity Cy-Lipo can mediate endocytosis designed nanovehicles into JEG-3 cells. The protein maintains its transportation function through circulation regeneration during endocytosis. Therefore, Cy-Lipo-based formulations showed tumor accumulation retention biodistribution studies. More importantly, DSPE-PEG 2k -Cy conjugation exhibited synergistic therapeutic effect choriocarcinoma. Finally, Cy-Lipo@MTX exerted an extremely powerful anti-choriocarcinoma with fewer side effects. This study suggests that overexpressed holds great potential as high-efficiency target rational design active targeting nanotherapeutics. Graphic abstract
